The opening (and ending!) credits to short-lived American comedy series' Sledge Hammer and Police Squad! as well as many, many fast-fleeting moments of Arrested Development (like whenever Buster appears with his bionic arm or the Bluthmobile is in action or Tobias wearing his Blue Man Group paint or first cousins' George Michael and Maybe's awkward sexually suggestive situations and lines.)

That's really hard. 
I mean somethings are absolute killer funny when you first see them, but once the joke's been sprung, subsequently they might not seem so funny as before. Other might have lasting laugh power but aren't THAT funny.

Examples.
Monica wearing a Turkey on her head in Friends
I split my gut at that one. Now it doesn't even bring a smile.

The boys farting around the campfire in Blazing Saddles
Makes me laugh every time, but not as hard as the turkey did that once.

I'm not sure I could pick a single favourite defining moment with so many great comedies I've seen over the years.
Think Faulty Towers, M*A*S*H*, Married With Children, Big Bang Theory, Precious Gemstones, The Office, Parks & Rec and yes...Monty Python

Fawlty Towers certainly has its own list

Guest '' This is the worst hotel I've ever stayed in'' , Major '' not having that , there's one in Paignton that's a lot worse!''

Basil to a guest who is disappointed with the view'' What do you expect to see out of a Torquay hotel window? The hanging gardens of Babylon? Herds of Wildebeast sweeping majestically across the plain?''

Basil waxing lyrical about the in growing toe nail that boaring itself into Cybil's toe

Thrashing the car, (had many of those moments myself!)

Cybil being lead into a cupboard on her birthday by Basil to avoid being seen by guests at her own party (they think she is ill and in bed as played by Polly)

Basils horse race winnings going from guest straight to Cybil, Basil then having a near break down over it.

Basil fawning over a crookster thinking he's a member of the aristocracy

The deer head falling on Basils head

Hiding the body of a dead guest

Manuel's rat suddenly appearing in a box of chocolates offered to a hotel inspector

Basil sending Manuel back into the kitchen that's on fire.

and many more I'm sure!

(Sadly ... the rest is not TV, but the imagination makes TV look like crap!)(And a lot of it was copied senselessly later ... specially the early Beatles films!)

Aside from that, there are a lot of images from THE GOONS that are way better than many of these films and their cartoonish covers. All three GOONS had been friends and mates in WW2 (some in Algeria - see Spike Milligan books!) and when they got together they put together a combination of surrealistic humor that defied your imagination.

"... fall down little kilt ... you can't stand up to my superior intelligence!" ... and it doesn't happen and he gets whacked.

Trains coming through the window. And by the time The Beatles sing she came through the window ... it wasn't even funny ... just clever copy! And The Beatles did a lot of humor that had Goon inclinations in their Christmas Shows.

Rooms upside down with the floor above you and the ceiling below you. Climbing Mt Everest from the inside. Various drug references that were likely psychedelic in nature as we would see later.

Six Charlies in Search of an Author ... where everyone takes a turn at the typewriter to change the story!

Various clever bank robberies including Neddie Seagoon running a bank and then losing his job. 

Several tea breaks ... specially in Africa. And while these days some might think they are not funny the BLACK jokes with Max Geldray and Ray Ellington. Funny as heck, specially the character "Throat".

  • "He's fallen in the water!" (Little Jim)
  • "You dirty, rotten swine, you! You have deaded me!" (Bluebottle)
  • "You can't get the wood, you know." (Henry, Minnie)
  • "You silly, twisted boy, you." (Grytpype-Thynne)

(Henry and Minnie are old folks barely walking and seeing anything!)
